Cytotoxic Effects of Peptides Fractionated from Bromelain Hydrolyzates of Soy - bean Protein

Abstract
To characterize the cytotoxic activities of peptides from soybean, defatted soybeans were hydrolyzed by various endopeptidases and extracted with solvents. Then the extracted peptides were tested for cytotoxic activities on several cancer cell lines. Ethanol fractions of bromelain-HSP (hydrolyzates of soybean protein) had significant cytotoxic activities on P388D1, a mouse lymphoma cell line. Bromelain-HSP ethanol extract was further fractionated by ultrafiltration into B1, B2, and B3 fractions. The cytotoxic assay revealed that the B1 (M.W.>10,000) fraction at 70% cytotoxicity had relatively potent activity. The B1 fraction was further fractionated into 13 fractions by reverse-phase HPLC. These subfractions were also tested on P388D1. The B1-12 fraction showed the strongest cytotoxic activity in cytotoxic assay. Thin layer chromatography and amino acid analysis indicated that this fraction comprised a glycopeptide which mainly composed of Asp, Glu, Pro, Gly, and Leu.
